#4945 NEPHROPROTECTIVE EFFECTS OF DIPEPTIDYL PEPTIDASE–IV INHIBITORS FROM PHENOLIC RICH FRACTION OF TRIGONELLA FOENUM IN DIABETIC NEPHROPATHY RATS

Abstract Background and Aims The most common effect of renal disease in the world is diabetic nephropathy (DN). Dipeptidyl peptidase–IV (DPP-IV) enzyme abundant present kidney, which increased nephropathy. A novel approach to treat type 2 diabetes mellitus (T2DM) along with DN, based on incretin hormones: glucagon-like peptide-1 (GLP-1) regulated by DPP-IV. We hypothesized that DPP-IV inhibitors isolated from phenolic rich fraction Trigonella foenum (TF) will regulate activity as well DN: in-vivo; in-silico kidney histology. Method TF high sucrose diet dexamethasone induced T2DM was explored in-vivo rat. Apart serum glucose, DPP-IV, inhibition activity, HbA1c, Insulin estimated. also examined GLP-1, albuminuria, antioxidant properties tissue Molecular docking (Gallic acid) Results High Dexamethasone administration (1 mg /kg BW 45 days) concentration cholesterol LPO increase scavenging free radicals generated oxidative stress, but after some time antioxidants such SOD, CAT, GSH decreased. However, TF, (71.1%), compared Sitagliptin (89.5%) significant reduction levels TC, TG GLP-1. Kidney histology showed change control. Isolated Gallic acid depicts conformer affinity energy – 5.3 distance RMSD i.b 38.669. depicted 8.9 3.826. Conclusion are nephroprotective, antidiabetic properties. lower blood glucose decreasing albuminuria increasing GLP-1
